Proteomics Facility

 

The Core Facility Mass Spectrometry, as part of the Bioanalytical Mass Spectrometry research group, offers a portfolio of protein analyses as a serceion a routine basis:

  • Molecular mass determination of intact proteins or oligonucleotides
  • Enzymatic digestion of proteins in-gel or in-solution
  • Separation of complex peptide mixtures by offline reversed phase (RP) HPLC at basic pH
  • Protein identification from complex mixtures by LC/MS/MS
  • relative and absolute protein quantification from complex mixtures by LC/MS/MS
  • Identification of (post-translational) modifications of individual proteins following gel electrophoresis or from solution, if necessary after enrichment of e.g. phosphopeptides

Our services include project consultancy, use of dedicated sample preparation protocols, mass spectrometric analysis, data analysis and finally communication of the results to the users. For data processing we employ a range of software solutions including Mascot (Matrixscience), Scaffold (Proteome Software), MaxQuant (MPI for Biochemistry), Proteome Discoverer (Thermo Fisher Scientific), Peaks (Bioinformatics Solutions), Skyline (University of Washington), DIA-NN (Charité) and Spectronaut (Biognosys).

We exchange methods and workflows on a regular basis with our laboratory at the University Medical Center Göttingen (UMG), which specializes in biomedical proteome analysis in e.g. tissues or liquid biopsies.
